A recent preprint abstract discusses the characterization of the endo-lysosomal ion channel hTPC2 and its role as a drug target, particularly for antiviral drugs. The study aimed to understand the mechanisms of ligand activation and ion selectivity in hTPC2. Through in silico simulations, the researchers found that hTPC2 adopts different structures depending on the presence of a specific lipid, PI(3,5)P2, and explained the conformational changes and ion permeation pathways. The simulations also confirmed that hTPC2 is more selective for sodium ions over calcium ions in the presence of PI(3,5)P2, and identified specific ion binding sites that facilitate efficient sodium ion permeation.
